Okay the # is a hash tag. Back when twitter didn't have an integrated search on the side bar it was very important to help sort out trends on the twitter search site. It's still used a little bit now, but less because you can search for specific words and trends can be hash-tagless.
There are a few that people seem to use the most #musicmonday where you type that somewhere in you tweet and link to a song you like or talk about music. I use this from time to time with my blip.fm posts.
The most popular one is #followfriday or #ff. You write @username for as many people as you want to pimp out, and #followfriday at the end. I wrote an extra little note that all the people in my post were from hdc. Sometimes I #ff horror bloggers, irl friends, etc. Other people see that tweet and check out those people, if they like what rthey see they can follow them.
You can do this for just about anything you want to start a trend with. For example: "omg have you seen the New Moon trailer yet? Edward is like so hot. #twilight"
